DIM (Diindolylmethane)

A broccoli-derived compound that modulates estrogen metabolism and activates anti-cancer pathways.

What it is

DIM (diindolylmethane) is formed when indole-3-carbinol (I3C) — found in cruciferous vegetables — is metabolised during digestion. It promotes conversion of estrogen from more potent forms (16α-hydroxyestrone) to less active, potentially protective metabolites (2-hydroxyestrone). It is fat-soluble and requires enhanced-delivery formulations for meaningful bioavailability.

Why it matters

DIM is relevant for hormone-sensitive conditions and as a broad anti-cancer agent. It activates AMPK, inhibits NF-κB, promotes apoptosis, and modulates the oestrogen metabolism balance. It is used in integrative oncology protocols for hormone-related cancers and as part of general anti-cancer supplementation.

The evidence

Clinical trials in cervical dysplasia, prostate cancer, and breast cancer show meaningful effects. University of California Berkeley researchers have published extensively on DIM's mechanism. BioResponse DIM (the most bioavailable proprietary formulation) has shown the most consistent clinical results at 150–300mg/day.

Dosing

150–300mg/day with food containing fat. BioResponse DIM (absorption-enhanced formulation) achieves significantly better bioavailability than standard DIM powder. Effects on oestrogen metabolite ratio are measurable via urine testing (2-OHE1/16-OHE1 ratio).

Cautions

Fat-soluble — requires dietary fat for absorption. May affect oestrogen levels — discuss with oncologist if on endocrine therapy for hormone-sensitive cancer. Standard DIM has very poor bioavailability — formulation is critical. Rarely causes temporary urine colour change (harmless).
